Molecules that bind to DNA by covalent or non-covalent intercalation, electrostatic interaction, and groove binding cause disruption in the double-stranded DNA structure. This situation is of great importance in studies conducted in the field of pharmacy to increase the effectiveness of antitumor and antiviral drugs, in DNA-based disease diagnosis and determination of toxicity properties of metal complexes. The purpose of this review is to evaluate the interactions of DNA-metal complexes and provide information about their determined biological activities.Öğe The effect of krill oil on Wnt/ß-catenin signaling pathway in acetaminopheninduced acute liver injury in mice(Walter De Gruyter Gmbh, 2023) Sahin, Yasar; Devrim, Alparslan Kadir; Alcigir, Mehmet Eray; Senol, Ali; Ekici, Husamettin; Devrim, Tuba; Sudagidan, MertObjectives: This study investigated the effect of krill oil (KO) on liver damage caused by acetaminophen (APAP). Methods: In the present study, the control and APAP groups were given distilled water by gavage for 14 days. In addition, the KO and APAP+KO groups were given 500 mg/kg krill oil by gavage for 14 days. At the end of 14 days, 0.9 % sodium chloride solution (saline solution) administration was applied intraperitoneally to the control and KO groups. Meanwhile, 220 mg/kg acetaminophen was administered to the APAP and APAP+KO groups. While some biochemical parameters in plasma were examined, some oxidative stress parameters in plasma and liver tissue were evaluated. Apoptotic and inflammatory responses of some primer sequences determined by quantitative Real-Time PCR (qPCR) in liver tissue. After histopathological examination of liver tissue, immunohistochemical analysis was performed with Wnt inhibitory factor-1 (Wif-1), beta-catenin (ss-Catenin), and 8-hydroxy-2'-deoxyguanosine (8-OHdG). Results: The Wif-1 positivity in hepatocytes increased significantly in the APAP group (5.29 +/- 0.71) compared to the control (1.14 +/- 0.51), and KO (2.14 +/- 0.55) groups (p<0.001). The 8-OHdG positivity in hepatocytes increased significantly in the APAP group (19.57 +/- 0.58) compared to the control (0.43 +/- 0.20), KO (3.57 +/- 0.48), and APAP+KO (4.00 +/- 2.53) groups (p<0.001). Conclusions: As a result, krill oil could be used as a nutritional supplement to protect the liver against acetaminopheninduced liver injury.Öğe Evaluation of Potential Candidate Genes in the Differentiation of Neoplastic and Non-neoplastic Conditions of Endometrium(Springer International Publishing, 2025) Devrim, Tuba; Devrim, Alparslan Kadir; Senol, Ali; Sudagidan, Mert; Atalay, Emine SumeyyeBackground: Endometrial cancer is one of the most common malignancies and alternative successful biomarkers are needed in the diagnostic process. Objectives: This study aimed to investigate candidate genes' diagnostic potential in endometrial neoplasia. Material and Methods: The expression levels of USP28, JAG2, AURKA, PGK1, HRPT1, EZH2, YAP, and P53 genes were evaluated by qPCR and/or immunohistochemistry analyses on endometrioid-type adenocarcinomas including all three grades, as well as from non-atypical hyperplasia, and atypical hyperplasia. Results: We determined significant (p < 0.05) increases in USP28, PGK1, EZH2, JAG2, AURKA, and YAP mRNA expressions in endometrial cancer tissues. Significant differences in the expressions of USP28 and P53 genes were determined between tumor grade groups (p = 0.002, and p = 0.005, respectively). Immunohistochemically, significant differences were found between the study groups (p < 0.001) and tumor grades (p = 0.013) by the evaluation of USP28. Statistically significant differences were found between all study groups (p < 0.001) and tumor grades (p = 0.008) in terms of PGK1 immunohistochemical expressions. A positive correlation (p = 0.002, r = 0.356) was found between p53 and PGK1. Conclusions: Considering our qPCR and immunohistochemistry results together, it was concluded that USP28, PGK1, EZH2, JAG2, AURKA, HPRT1and YAP expressions could offer beneficial expression values in precancerous lesions and endometrial cancer. © The Author(s) 2025.Öğe Impact of melamine exposure on apoptotic proteins and oxidative stress markers in mouse hepatic tissue(Ankara Üniversitesi, 2025) Devrim, Alparslan Kadir; Devrim, Tuba; Ekici, Hüsamettin; Sözmen, Mahmut; Şenol, Ali; Kuruca, Nilüfer; Sudağıdan, MertMost melamine studies have focused on renal toxicity and its effects on the liver are still not well known. Despite the progress made in the understanding of cancer mechanisms, the strategies based on research are still at an incipient stage and the conventional treatments, mainly chemotherapy, remain the most widespread form of cancer treatment. However, these conventional therapies have several drawbacks, such as off-target effects, drug resistance, toxicity, and poor bio-availability of drugs to tumor tissue. Smart drugs and nanostructured drug delivery vehicles have been generally applied for cancer therapy due to improvements in medicine and technology to solve these such limitations of cancer treatment. Therefore, smart drugs offer more effective and less side-effect treatment for cancer compared to conventional therapies. In this review, we aimed to examine smart drugs, which have been increasingly used in cancer treatment in recent years.
